Do you have the automatic yum doodad (whose name I can't remember offhand) running as a daemon?

Are you sure you're root?
Also, possibly there are stale locks. The locks can be removed with `rm -f /var/lib/rpm/__db*`, but it is safer to reboot, which will remove the locks at a time when it is certainly safe. And the advice to use lsof first is good.

Have you got the software updater or software installer running? they will lock the rpm database until you close them
