Managed to reproduce the error and have upstreamed it here:
http://bugs.locamotion.org/show_bug.cgi?id=1505

The moz2po command needs a target output directory.

If you ran:
  moz2po -P en-US pot

Or
  moz2po -t en-US po-xx xx

Then this command would have worked.
