You should do:

Help -> Software Updates -> Find and Install ...
Search for new Features to Install -> Next -> New Remote Site ...
Add http://subclipse.tigris.org/update_1.0.x and click finish
Now select Subclipse and you should be on your way to having subclipse
installed.

Actually adding an Extension Location should work if you had an Eclipse
Extension. AFAIK, subclipse only releases an update site. If it doesn't
work with an Eclipse Extension, please file a bug.
