problem in squirrellmail or sendmail

I test my sendmail is working  for this  way


[root at bigboy tmp]# telnet mail.my-site.com 25

Testing sendmail interactively

This mean my sendmail is working   and after this i check file in
/var/spool/mail/userfile

i see mail whose i send.

Now I try through Mail client (Squirrelmail) then
I am anable to send mail.
